The Argentine has been suffering from physical problems and he was an unused substitute for his country against Venezuela this week.
It is unclear whether he will be fit for the next game against Bolivia which suggests that he could be doubtful to face the Bees as well.
Martinez did not come off the bench against Aston Villa before the international break and it may have been for the same reason.
United have had a dreadful start to the campaign and they are currently 14th in the league standings with only eight points from seven games.
Erik ten Hag is under huge pressure after the poor run of results and he could be sacked if United can't beat Brentford after the break.
