He suffered the setback in the second minute against Real Madrid this week, and United have now confirmed that the issue is serious.
The youngster is set to miss the first part of the forthcoming season. It is a blow for Mainoo, who was appearing in splendid touch.
With his setback, United are likely to block the departure of Scott McTominay, who has been touted to head for the exit door.
The club are currently looking to land another defensive midfielder in Sofyan Amrabat, but are still keen to keep McTominay.
Erik ten Hag wants quality competition and depth in every department, and McTominay may have to wait until January to leave.
While McTominay is likely to stay, United are widely expected to part ways with the likes of Fred and Donny van de Beek.
